If
you double click on it it should play the sound that will play when that new mail comes in. The actual name of the file to be used is located in the box to the left of the Test button. If 'New Mail Notification' is not there you can try resetting the sound scheme (drop down list near top) to windows default.

Somedays I wish I'd use a newsreader. Vista gave me fits when I tried it. It didn't see my video card at all and my webcam doesn't work with it. And they removed Fax Services! The bastiches.
